Texas State University, San Marcos, Texas.https://hdl.handle.net/10877/15951The ability of guanine-rich DNA to adopt various folded forms is well-established and these G-quadruplex structures are likely involved in telomere maintenance. The folded structures are stabilized in solution by binding of cations such as Na+ and certain small molecule ligands. One such ligand, TMPyP4, has previously been demonstrated to interfere with telomeric DNA processing by binding to G-quadruplexes in the DNA. However, for the G-quadruplex forming oligonucleotide, Tel22 (based on the human telomeric DNA sequence) TMPyP4 significantly decreases the stability of the folded structure in the presence of varying levels of Na+ ion.
